When should you start looking for a job if you are graduating soon and you are taking a year off between undergrad and med school?

You should probably start searching now then start applying as you get closer to graduation.

you should be applying (not just looking) now. especially if you're looking for jobs at a university or hospital. it can (and often does) take at least 3 months to actually get hired after you submit the applications.
