Does it make much difference when responding to an interview invite to phone immediately or just send an email with my preferences for date options? Most likely they would be sending the confirmation via email so does it make much difference?

Sometimes responding by phone gives you a better shot at securing the date you want.
 
If you are asking, then just don't bother. Neither is better. If the person is on the phone, the call was wasted. If they don't read their email frequently, the email was wasted. Once I called a program to get a voice mail stating that person would be out of the office all week. I emailed my request, and got a reply in 15 minutes.
