Km and Vmax

Hi guys,

For a single enzyme and it's substrate, would the Km and Vmax always be the same? When would it be different (excluding inhibitors)?

I don't understand, Km and Vmax are completely different variables.

He's asking would the Vmax and Km remain constant across different reaction conditions.

Well, Vmax would change depending on the concentration of the enzyme.

Across a single reaction, yes, but Vmax is just Kcat times the total enzyme concentration, so for a given enzyme/substrate pair Vmax can vary between different reaction conditions.
