Do you have someone in mind to write the letter for you? I am confused in what you are asking.
 
How does one go about getting a LOR from a senior university if he/she transferred from a JUCO!?

Contact the individual and ask if you can meet with them. When you meet with them provide a CV, Personal Statement and whatever else they may need to write you a letter.

In other words, just the same as you would otherwise.

This seems a bit unclear, did you start out at a 4 year university and transfer to a JrCO or the other way around? Basically if you know this prof enough to write an LOR just go and ask them, you do not have to be a current student to get an LOR as long as the prof still knows who you are.
 
I'm starting at a JUCO transferring to a senior university soon. And I'm wondering do I ask one of my professors from my juco to write a letter or just wait until I can get one from my one of my senior university professors?

I don't really get what you asking but why not get both? If you are asking which one, choose the LOR with which you know the Professor the best.
 
Did you have to transfer from a Juco to a university? If not then it can be hard to understand. So basically I'm asking how do I get a professor to get to know me well enough to write a LOR?

no i didn't transfer. I would recommend going to office hours and/or joining a research lab.
