Here @ the MCC (Molecular Cloning Club), we have a few simple rules that will save you the agony of failure. I'll post the first one and feel free to add your own.

The first rule of Molecular Cloning Club:

Never accept someone's word that the sequence is "good to go." Always sequence your DNA before hand. Otherwise two months of work will go down the drain.

It seems that even PI's aren't immune to handing out bad template DNA.

I would like to suggest a boycott of nucleic acids. This will avoid all future frustrations with regards to cloning as well as various assays (eg southern's, northern's,PCRs of all flavors).

I always created an anti-sense templates for back up as well, then rt pcr away. Reverse transcriptase seemed to be more efficent than DNA poly taq
