I know most schools require 2 science and 1 non-science, but plenty of schools are now lenient as to the type. For the schools that say they need just one science (and the others can be from anyone else) or schools that are fully lenient as to the type, is there still unspoken requirement or expectation that you send in 2 science and 1 non-science?

I believe if they don't specify, you send whatever letters you have that you believe are the strongest. To be safe, the basic set of letters I would get would be 2 science, 1 non-science, and try to get them from upper class professors since some schools specify upper class. With this, you'll be set for most schools. Get one more letter from a PI, doctor you shadowed, or volunteer coordinator, etc, and you should be set for schools that want a letter from a MD or person who knows you outside of academics.
