Post-Secondary Letter of Rec Submission

Hey all,
I want to send in a LoR from my current employer to a few schools I was placed on hold for and when I called each of the offices of admission on how to submit them I got 50/50 responses saying I should either submit via AMCAS or just use one of my updates. What's the difference (if any) between these methods? I'm hesitant to go through AMCAS because I want for them to see this is a post-secondary letter and not mess up my spot in line (if that's possible), but if I just upload the letter using one of my updates do they care that I could have potentially read the letter?

Also wondering about this. I have a new LOR from someone with strong connections to a specific medical school (alum + research center named after them), and I would only submit the letter to that one school. Is it worth submitting at this point? It would likely be pre-II if that matters.
