Seeking Advice: Waiting To Submit Secondaries Due To LOR Missing?

Hey all. Essentially, I have one LOR writer who has yet to submit their letter to Interfolio. I believe it will be a strong letter, so I for sure want schools to see it when they read my application. Unfortunately, this person is on vacation, and will not be able to submit the letter until likely around June 27-28. Then, I will have to send it from Interfolio to AMCAS. In the past, this process has taken up to a week.

So, my question is this:
Do schools send out secondaries on June 27 when the first batch of applications get to them, and if so, should I wait a week or more to send them back to ensure my LOR is available? I don't want to wait too long, but also want the letter to be read when they first review my application.

Do your secondaries as they come in, because this is something under your control.
Letters can come in later and are reviewed later by schools
 
How many letters are already in? Is this a critical science letter you "must" have to fulfill schools' letter expectations?

The screeners don't always read the letters; often, we just check to see if you have the minimums without reading it too deeply.

Currently, I have 4 letters in. These have all been "sent" to schools (with the exception of USC - only sent 3). It is not a critical requirement, as I do have letters already from a PI, physician, and 2 science professors. Thanks for the response!
