I have never shadowed a DO before, but shadowed MD. If I apply to DO schools, would they care if I shadowed a DO or not?

Some do, like OSU-COM.

If many others have, do you really want to handicap yourself by not?

Probably not, but check with the schools you are applying to. All the schools that I had applied to didn't really care.
 
I'd imagine it would be worth the time and effort, as it probably gives you a leg-up and something to talk about during the interviews...

I shadowed a DO myself, and at every interview I was at they asked me if I shadowed a doctor and what kind. I have heard from a few friends who did an MD shadow instead that they got a lot more questions about how serious they were about going DO vs. MD.
 
Not all school require a letter from DO, but they would like to see applicants to have exposure to Osteopathic Medicine and/or DO.
