Yes, there are some fundamental differences between the two - physics being one 🙂 . If you would like to judge for yourself how they are different and how prepared you are for the various sections then take a look at Kaplan's DAT prep book. Perhaps the most noticable difference is the PAT (perceptual ability section) portion. It's something few people can just ace on their first try and most people need lots of practice time to do well on it.
Also, unlike the MCAT, the DAT normally takes between four to four and half hours to complete as oppossed to a whole day for the MCAT. Plus, you can sign up to take the DAT just about any day of the week and not have to worry about being pinned down to just two or three dates out of the year.
