Timing and Strategy for Sending Update/Letter of Intent for Post II Waitlist

If I was placed on Post II waitlist for a DO school back in Ocotber. When would be the best time to send an update and letter of intent if the school is my top choice? Additionally, is it fine to send a letter of intent and activities update in the same letter or better to send them separately?

(The school did mention that they accept both at the interview)

Updates, if substantive, would be good now. An LOI I would wait to March or April as people start to decline offers.
