I am doing some passages on TPRH bio and some of the advance passages are putting a big blow into my confiddence. For instance, passage 12 in bio, I read it and I fail to understand most of of it. Consequently, I miss 4 out of the 7 question on it. Even the explanation provided by TPRH went way over my head.
Can someone who has TPRH explain answers #2, 4, 7 bio passage 12 ?
 
You need to look up how to read plasmid maps. There are a couple youtube videos. See how they get spliced. (the circle thing is getting spliced by the endonucleases at sites marked on the circle) - will help with all 3 of your questions. Also for 4 - you need to know how once it's cleaved, what is recombinant DNA. And the kb info is on the second figure.

The key is knowing how to read the map, what endonucleases do to it, and how recombinant plasmid/dna is created.

That is the point/concept TPRH is testing you on there. The questions you missed were based on understanding those concepts.
 
Also this is a very important concept to understand for the experimental passages of the MCAT. This is a very common process and needs to be understood.
