Unsigned letter in committee packet

I submitted my committee letter packet with my AMCAS last month. However, I recently found out that one my letters was unsigned, and got my professor to sign his letter. My question is, should I resubmit this letter through AMCAS, or send it directly to the schools that I am applying to? Some of my schools have already downloaded my committee letter (with the one unsigned letter).

The thing is, I only found out that one of my letters was unsigned because a medical school contacted me, which is why I'm worried that other medical schools might do the same.
