So I’ve had 3 interviews, but I have been waitlisted at the first one, deferred at next (basically waitlist), and I haven’t heard back from the 3rd.

Would my chances of receiving an acceptance increase if I sent each school an additional LOR from my current boss at my clinical job? It was my most significant premed activity and it would be a really strong letter.

It is unlikely to help at this point. Most schools have made the offers it will make (aside from the one school that hasn't gotten back to you post-interview-- that could be the one!) and those schools are waiting to see how things shake out before they make more offers. They may have just enough students and not need to go to the waitlist or they may have too many take their offers, a rare and horrible situation for schools. In those cases, a LOR will not get you off the waitlist because there is just no room for you.

If a school has a spot and will go to the waitlist, some schools will have a numbered waitlist and you won't be chosen until they go down the list in order and reach your name. A letter will not help. Some schools will use the waitlist to balance an out of balance class ... seeking more men (or women), more rural (or urban) students, or otherwise balancing a class that might have been off-balance due to an out of the ordinary pattern of offers being accepted. If you aren't what the school needs to restore "balance" to its class, then you won't get off the list and a letter won't help.
