I just received an update letter from my supervisor with which I'd like to update my current file. Is the best way to hav the individual upload the document to interfolio and then to have that service send it to AMCAS? Then to have AMCAS to send it to my schools again? I'm not sure how I add additional letters of recommendation to schools I've already submitted my AMCAS to.

i think it's best to contact AMCAS and ask if you can add new letters. if so then have your professor send the letter of recommendation to interfolio which will then send it to AMCAS, and AMCAS can then take it over from there like you mentioned.

Unfortunately I'll be at work all day and unable to leisurely use my free time to call AMCAS (it takes way too long to get through to them). Does anyone have experience sending a letter to a school through amcas having already submitted their application to them?

Normally you just create another letter entry and add it to the schools you want it sent to. Considering it's 10/15, though (and I think that's when AMCAS "closes?") I'm not sure if it's that simple.
