Can this person be the PI? Maybe in the world of clinical research / clinical trials this is different, but I've done translational/basic science research where I worked with a postdoc but also in some cases where I worked directly with the PI (the lab was something like 10 or so people). Would I be obligated to list my postdoc in the first case, and could I list my PI in the second case? Or can I list the PI in both cases?

It can be the PI if you worked, say "half your time," with the PI. If you barely/never saw the PI, I would be more inclined to put the postdoc. It seems like you are fine to put down the PI in your case though.
