I’ve heard mixed things. There are people I know who have sent schools letters of interest and later received interviews, and there are also people I know who’ve sent letters and not heard back. With that being said, sending a school an email that you’re interested in their program only takes a few minutes, and it can only help you.

To add to this, you have nothing to lose when you send a letter of interest. It will only help strengthen your chances of getting in.

this is 100% false. the only thing it does it make you look like a complete idiot. your "letter of interest" is YOUR APPLICATION.
 
According to Adcoms in pre-med forums letter of interest screams desperate - they know you are interested since you applied there.
 
If letter of intent is what you desire, then do not let it cause you to perspire
 
IMHO, i think letters of interest should be reserved for when you are put on a waitlist after your interview to express your continued interest in the school.
