I was waitlisted at four schools and I received an acceptance from one of them a little while ago. My top choice school still has me on their waitlist. The acceptance offer only lasts two weeks. If I accept the offer, does this take me off of the waitlist at my top choice, or is it possible to hold a place in the class and wait to hear from the other waitlisted schools prior to the orientation?

You can accept the offer and remain on the waitlist at your top choice. You will only need to withdraw from all waitlists when you matriculate to medical school (i.e. during orientation).
 
Word of advice! Let the other school(s) know you're still interested. They can pass over you if you have an acceptance and haven't shown much interest since you already have an acceptance. Some schools don't care and go down a list but its better to be safe than sorry.
