I'm a biomedical engineering student who's working on an anesthesia machine graduation project, in my research, I found that every drug has its own concentration and dose which depends on patient's weight. My question is, if I want to anesthetize a patient's hand, should I measure the weight of his hand and put it into the equation?

No. You'll likely be doing a nerve block and you'll use fairly standard injection amounts of anesthetic (lidocaine, marcaine, etc).
