I am trying write a CV for dental school. Are mission trips counted as Volunteer Activities in application/CV? How do I include my time commitments? Before each mission trip, I spent about 100 hours preparing for mission trips.

Yes, it is considered volunteering. You could put time commitment in a bullet point with the other details of your contributions.
