I personally like #1 for its non-academic theme. I wouldn't go with #3.

Hello, I'm struggling to find a good topic for a challenge on your journey to medical school secondary essay. I would appreciate any feedback. Thanks in advance!

I am considering

#1 ) Taking about how I was a competitive runner in high school but not making the cross country team in college. I could elaborate on how this was a mental challenge that I overcame.

#2) Talking about having my first two research grants rejected. Then succeeding in securing funding and winning an award for the project.

#3) Talking about being rejected from medical school.

1 all the way. 2 and 3 make you look like the typical hyperachiever/lab rat who has no real life.
