With a few exceptions (I think Georgetown likes getting letters of continued interest from the people they waitlist for example) letters simply expressing interest with no update included are going to have no effect at all

It depends on the school. Some schools like Georgetown and Jefferson are needy and love to watch applicants grovel (as @Goro likes to say). Other schools don't care and view them as fluff.
