thats along the lines of what i was thinking but was not sure.

Lane 1: The protein with signal peptide
Lane 2: The protease cleaves the protein completely.
Lane 3: The protease cleaves the protein completely and the detergent dissolves the microsomes.
Lane 4: The protein without the signal peptide, thus migrates further down.
Lane 5: The protein without the signal peptide, and it undergoes post-translational modification (PTM) that protects the protein from being cleaved by protease.
Lane 6: The detergent dissolves the microsome, thus no PTM for the protein. Eventually, the protein gets cleaved.
