in galactosemia, the polyol which is formed, galactilol caused toxicity in the lens due to an osmotic pull of the lens, with swelling and cataract formation, my question is, when galactose 1 phosphate accumulated in the liver and brain, how does it cause toxicity??

I think it's because the formation of gal6p is tying up all the phosphate, so you don't have the phosphate needed for ATP production and other energy processes, leading to cell death.
