How many letters of rec needed?

I know that most med schools required 3 LOR (2 from science and one non-science). However, I have done biochem research and was wondering which of the following combos I should choose:

4 Letters total - 2 from science, , and one from my PI

OR

3 letters total - 2 science, and one english prof

OR

3 letters total - 2 science and one PI

I like option 1. It seems like the most well-rounded.
 
I think if you add (1) physician LOR it would help
 
I would get 6 in total:

3 science profs (1 physics, 1 bio, 1 chemistry) - some schools are specific about having at least 1 bio or 2 physical science professors so 2 chem instead of 1 physics and 1 chem work too.
1 non-science prof (english/econ/psych/etc.)
1 research
1 person who knows your character but still has some pull (a physician/pre-health advisor/etc.)

I applied to a lot of schools and I found that this was the best combination.
