How strict are letter of recommendation requirements?

Hi all, I'm a non-trad career changer currently working in drug discovery R&D. I have one science letter from undergrad, and two letters from my current and previous boss. I am trying to obtain a letter from my graduate program but I have been largely unsuccessful. My question is, how strict are the science/non-science professor letter requirements? My bosses can effectively serve those roles and write stronger letters about my abilities than a professor could, so I'm wondering if schools will understand this as an exemption.
