How to ask this question?

Do the latter. Send a SHORT email explaining who you are, what your research background is, and why you're interested in their lab. Short as in a couple sentences for each of those three. Attach a resume. Don't include a letter unless they ask for it. Definitely don't ask the likelihood they'll hire you.
 
Ask them in person if possible or take one of their classes first so they can get some kind of idea what type of student you are.
 
Take prof's class, nail said class, then ask about open research spots.

This. Also consider attaching a transcript so that he/she will know what skills you should have at a quick glance. If you're having a hard time with replies, follow up with your professors. I just had more than a week of silence from a professor that was writing a letter of recommendation that was due today, despite multiple emails reminding him about the due date.
 
Departments at my school often have get togethers that students are invited to attend. If yours has one too go and see if the professors are there
