Hi! I was wondering if it would be okay to submit a new letter of rec in the coming weeks. Should I upload to AMCAS and send notice in an update letter as well? I have all my requirements met but this is a strong MD letter from a prominent figure.

No, I just thought it could bolster my application. All my rec letter requirements have been met, just didn't know if this additional letter could help since it would be my first MD letter and they are well-regarded in their field. Not prompted @Mr.Smile12

It is unlikely to move the needle. Admissions will thank you if you send less letters their way. If this is someone you worked with extensively (not shadowing), then a research or employer letter from an MD has some merit. Otherwise, an MD letter is unnecessary.

Also I have a couple of updates to my application (accepted pub, pub in peer review, new QI project) that I wanted to send to my schools pre-II.

Should I wait until the paper is published and I have a doi (which might take an additional 2-3 weeks), or is an accepted manuscript enough to update now instead of the wait? Trying to balance since it seems like it's mid-cycle now.

Wait and send it in a month about the publication. You are free to bring it up ahead of time in any interviews if there is an appropriate opportunity.
