When schools say they require a letter of recommendation from 2 science faculty does that mean the faculty must have been your teacher? For example I know a science faculty outside of the class room setting and he/she is willing to write me a nice letter. Can I list him/her as a science faculty letter writer?

Would it work when they specify that it has to be a science letter from an academic setting?

2 science professors that taught you in a classroom setting, not a tutor etc..so in your case the answer is NO.

If I provide a transcript I think they would be able to discuss my academic ability to some degree.

I think so too! But I would do this as a last resort. Like unless you REALLY don't have two other science professors who taught you to write one for you.

I have 1 from a science prof who I've taken several classes with and 1 that I have not taken any classes with but have done research under. my prehealth adviser said it's fine, but I haven't applied yet so idk.

So if you are taking a class that is taught by multiple professors, can you ask 2 different professors from that same class?

Wh wouldn't it be the best strategy?

The point of a LOR is for the medical admission committee to get a better feeling for who you are beyond the numbers. That's why they ask for so many LORs. If you ask professors who taught the same class you took, they'll likely have the same things to say about you (unless one of them has taught another one of your courses or is your PI). "So-and-so worked really hard in my class," etc. You want to get as many different perspectives on your character as possible.
