Are we expected to know the Lineweaver Burke plot for the new MCAT? The Khan website includes this topic, but I heard that Khan actually includes some topics that are not on the MCAT and I was wondering if this is one of them.

You are expected to know enzyme kinetics, specifically Michaelis-Menten kinetics - check out page 12 of the new AAMC outline.
A good additional resource (outside of whatever books you may have + Khan), I'd suggest checking out this website as it's been pretty helpful:

Although it doesn't look like the Burke plot is a topic on the content outlines, it is just an off-shoot of MM kinetics, so I'd say it couldn't hurt to know it.

Ooh, I see! I actually did go though the AAMC outline that you helped me find in my other thread but I'm not familiar enough with biochem content itself that I wasn't exactly sure where Lineweaver Burke plot fit into the whole picture. Thank you!
