Can you make any suggestions for turning a pre-interview hold into an interview invitation? I have sent my one letter of update. I have considered that if I send any more I might be viewed as a pest.

I think the letter is really all you can do. I disagree with the thoughts that it's risky. How would it be risky? Would they read it an decide they hate you and throw it out? No. The worst that happens is they ignore it, unless they are HUGE a-holes. The best is they respect your interest in the school and are impressed by whatever you're doing.

It's risky because some schools truly are not receptive to such letters before an interview, or at any point in time. At other schools, they will take it into consideration. But it's difficult to know which schools are okay with such a letter, and which aren't. You want to seem like an interested and eager candidate, but not a desperate one.

Why is it risky? If the school doesn't accept pre-interview letters, then the worst they could do is not accept the it. But that would mean that the office of admissions would dismiss the letter, and it wouldn't ever make it to the ADCOM.
Would it really affect the decision made on your application, if the ADCOM never actually sees/knows about the letter?
?? It's a simple matter of asking the admissions office if they accept update letters.

Most of the schools I emailed said that sending a letter was no problem. I was told by multiple places that I can update my application in AACOMAS (not sure if AMCAS is the same) and that they would see the updates. They said I could also send an email to admissions detailing the update, and anything else I wanted to say.
OP, if you've already sent 1 letter, I would wait at least 1-2 months to send another. If the first one doesn't do anything, likely the second won't do anything either, HOWEVER... I do know of people being pulled off waitlists (for acceptance) by sending multiple letters (like 1 every week-10 days....) That said, best of luck, and at worst the letter does nothing, at best it helps, I highly doubt an admissions person is going to have you rejected for sending an LOI
