Hi,
A High School Jr would like apply for 6/7year BS/MD program in coming Aug/Sept 2010. Does he needs to register to AMCAS ?

No. The application should be available through the school/program. BS/MD programs do not use AMCAS, so a separate application will need to be filled out for each program he applies to. If I recall correctly, they are usually available in the early fall.
