I know that SDS Reducing PAGE disrupts Quaternary structure and Secondary Structure and that SDS NON-reducing PAGE only disrupts quaternary structure.

My question is: if the question just mentions SDS PAGE, do we assume reducing conditions? I have encountered a problem on the Section Bank where this was not clarified.

Standard SDS-PAGE does use reductant. When you run an SDS-PAGE, you usually reduce the disulfides before because you want to separate based on weight - that's why almost every gel is run with a ladder. Usually they'll only specifically specify if it's non-reducing.
