If I currently have 4 LORs (2 science, 1 nonscience, and 1 from my graduate school advisor), is it advisable to assign all four to medical schools, or rather just send the required three they want? Would they even read all four?

Which letters you want to send depends on the school. This is why you can choose which letters to send to which school on the AMCAS. Each school has its own letter policy. If you can't find it on their websites, e-mail and ask them what they recommend. Many allow extra letters to be sent and will read them. I think the max is like 6. So look thoroughly into what each school you apply to wants/accepts and act accordingly.

Also, do you have any letters from an employer/supervisor (whether medical or not) of some kind? It's good to have a letter from someone not involved with your academic life so they can hear about who you are outside of school. The best is obviously a doctor or other medical professional you shadowed or volunteered with, but really anyone who knew you professionally outside of school is good.

Depends on the school - some schools have a maximum limit of 3 letters, some will accept many more. Don't assign your letters until you get your secondary applications because the school's LOR requirements usually accompany the secondary app.
