I'm a 3rd year in college. I have a fairly high mcat and gpa already with lors going out in late may early june, but I have my 3rd biology lab in progress this quarter (molecular biology, spring 2013). It will remain as "in progress" on my transcript until June 24th. Should I wait for that transcript and the verification then? or should I submit my current transcript and the AMCAS ASAP (June 1-2) to get secondaries ASAP and just update the AMCAS later once the the new grades gets posted?

You can't update AMCAS after it's been submitted for verification. If your GPA is high enough (and this new grade wouldn't increase your BCPM or your cGPA significantly), then I would just submit and forget about the grade. Being verified before schools start sending secondaries (end of June/early July) is great (though by no means necessary)!
 
Once transcripts are submitted and verified with your app, new transcripts won't be accepted and grades won't be changed. The only way you could send new grades in is essentially if the school you apply to asks for it or through update letters.

However, I wouldn't wait. I have like 4 class this spring quarter and I'm just going to go ahead and turn in my app. You won't get the verification process started until your transcripts are received, which is take a week probably. That puts you in July, where you'll probably have to wait longer to get fully verified (which is LONG for July applicants). So not worth the risk.
