Ok, so I won't be done with my classes until June 8th, which means my grades won't be in until June 10th.

I am confused as to whether I can submit my application on June 1st or do I have to wait until my semester grades are in?

I was planning on submitting my AMCAS on June 1st, and process my transcripts on June 10th. Would this make a difference as to how fast my AMCAS will be processed?

The process of matching an official school transcript with what you list on your AMCAS is the process known as verification, and it is the time limiting factor for early submissions. Generally speaking, if you submit your application after your transcripts are in by June 1st, you can expect to be verified within 1-7 days which will mean that you will be verified right when receiving secondaries.

However, if you choose to wait for your grades, you can expect AMCAS to receive your transcript (which is hopefully available around June 10th) a bit later (towards the end of the month), and then get verified sometime in July.

You do have a choice: You can 1) wait for your grades to come out and send a complete transcript, or 2) submit your transcripts missing the last semester/quarter grades and put the classes you are taking now as "currently enrolled". I would recommend you do the second option if your GPA will not significantly be impacted positively by the oncoming grades, as early verification is generally the most precious factor you have in your control at this point.
 
I'm a senior in basically the same situation. So if I do the second option and get verified on June 1st without spring grades should I have a transcript request into the registrar to send final grades to AMCAS once they are available or at that point should I just wait to send an updated transcript once the rush is over?

That's a dilemma. My Spring semester grades will be in by May 15th. However, I am planning on taking a class during May term. The difference would be from a 3.72 to a 3.75...which I think is slightly significant as I am on the lower end of the GPA spectrum.

What if I sent in my transcripts with my 3.72 and then sent in my May term grades on June 10th? Would AMCAS update my May term grades then?

If it was me, I wouldn't sacrifice getting verified several weeks later for just a 0.03 GPA difference... I would send in my transcript as soon as AMCAS opens. When you do get your final transcripts, AMCAS is not going to update your grades for you. Instead, you can update schools individually on your recent grades (if they are good) later in the cycle when you are waiting for interviews, which gives you something to talk about should you decide to write update letters/letters of interest.

I would just update the schools with the new transcript
