I think they usually take it as someone being desperate to get into a school than anything else. If you can do something update worthy and send an update with a letter of interest that may work.

Think they don't really mean anything because of course you would like to get into the school. I do think that they get a little more meaning if you have other acceptances that you'd be willing to give up and you communicate that in the letter but in the end, it really depends on the school
