When is the best time to do audition rotations?

Is there a window where its the only time this can be done? Do you absolutely have to do an audition rotation to score a spot in a program for a competitive residency (DO or MD)? Is it very difficult (or impossible) to get into a DO or MD program in say, a University of California San Francisco or Stanford program?
 
Usually audition rotations are done early in 4th year. You want to have time to get your LOR to submit with your application and generally you want to apply early.

You don't have to do an audition rotation to land a spot in a competitive program but some programs only interview those that rotated through.

Good question, I was curious about Mayo (although FL does have a Mayo clinic, too), and I was also curious about some schools like Columbia too.

What is the difference b/w FL's Mayo Clinic and Minn's Mayo Clinic? I know the original one is located in Minn.; however, is there a big difference in the quality of the attendees and residents and the programs avail b/w the 2?

I don't think program websites would explicitly state that they have a preference for MD/PhD students, MD students, students who rotate through their program, ivy league schools, etc. These trends are discussed in the residency forums in the link above.
