How vital is it to understand genetic technology like blotting, and stuff like PCR, RFLPs.
 
Just know the bare basics

here are some basic lab techniques you should know.
PCR:
Western blot:
Southern Blot:
Northern blot:
SDS PAGE
DNA microarray technology
Gram staining

here are some more advance biochem techniques you might not need to know
bradford assay
ELIZA
benedicts test
 
I'd know these:

PCR - DNA multiplication (know all the denaturing, annealing steps).

Gel electrophoresis - Separates by size, attracts nucleic acids down an agarose gel with positive current.

Size exclusion chromatography - Separates by size, larger molecules travel faster than smaller.

DNA Microarray - Measures different gene expression levels simultaneously. Uses "probes".

For blotting: Memorize SNOW DROP

Southern - DNA
Northern - RNA

Western - Protein
 
I have a whole santa bag full of mnemonics, but most of them aren't going to make sense like SNOWDROP.

A good one to know is SEVENUP

"Large Random M & M" for Hardy-Weinberg:
