I've been working with a postdoc in lab. Can he write me a letter?

Yes, and it's common that the PI would be willing to cosign it and lend his "name" and academic credentials to making the letter stronger.

Like catalystik said, it would be better if you can get the PI to be the main name but work with the postdoc to craft a strong letter.
