I think number two sounds the strongest. Or number three. For reasons you mentioned, I would not go with number one.

2 and 3 sound most interesting to me. possible to combine

I'd suggest combining 1 and 2. Your number 3 is too common without more elaboration, many people have siblings and can speak to their influence on their life or character. A diverse experience with a sibling could include: living with a sibling that has challenging disabilities, sibling with destructive behavior (gambling, alcohol), etc.
