In the eyes of the admission's committee? Assuming both the quality of each letter are the same. Would that extra DO letter from another physician help?

Absolutely multiple DO letters would be beneficial! This means you have experience with two separate DO's and a likely more diverse understanding of osteopathic medicine. This may actually separate you a bit from other applicants in areas where there aren't that many DO's practicing.
