grade forgiveness

How tolerant are the Big 4 schools with respect to grade forgiveness in your early years? If say you started off horribly and ended very strong but your overall average is still <3.0.
Would a reasonable MCAT (high 20s/low 30s) and a very high grad GPA compensate?
