I just realized that one of the professors who wrote an LOR for me did not include a letterhead on it. I am having trouble getting back in contact with him. Is this a deal breaker, or will I be alright?

Assuming that it is one which is needed to meet the school's requirements, then yes it is vital. Required letters without letterhead or a signature won't be accepted, but additional letters (not meeting school's requirements) don't need to be on letterhead.

I thought the consensus on here is that not having a letter head on a LOR isn't that big of a deal. I mean a DO I shadow for example doesn't have a letterhead so my LOR from him/her has a makeshift letterhead. I think you'll be fine OP. I'd imagine most schools are okay with it. If not, they will contact you.

I contacted almost 15 schools regarding this since I was getting an additional letter from somebody who retired and didn't have access to a letterhead. School's said they don't mind for additional letters, but would definitely bring your letter into question from a faculty letter w/o letterhead. I would try and figure out a way now just to be safe than wait and have your application delayed.

I guess it depends on what you consider a letterhead right? I mean OP could make his writer a letterhead and just tack it onto the letter. It doesn't have to be that complicated. If they dont have a letterhead they dont have a letterhead. It's not like I'm going to make my letter writer CREATE a letterhead just for me.

In your case OP, since it is faculty, I would do my best to get an official letterhead on that letter.

One of my science LORs (i.e, certainly not an "additional" letter by any means) was not on a letterhead - I found out because two schools (out of over 20 that I applied to) contacted me to let me know they could not accept the letter without letterhead. All other schools seemingly had no issues with it - I was offered many interviews and was accepted everywhere I interviewed. Worth noting, at my top choice school, I only submitted two letters - the un-letterheaded science one and my likely average DO letter. Still was accepted, so can't imagine this was actually a big deal. YMMV but certainly, don't panic, especially if you can't get in contact with your letter writer.
