I have already asked my professors to upload my letters of rec. to two other programs, I want to ask to send them to one more program. Do you guys think that's asking too much?

Do you have interfolio? They would only have to upload once and most programs accept interfolio.

are you using aacomas? Why are they sending letters outside of aacomas?

I want to apply to multiple professional programs, should I tell them about why or just request one? I suspect that they will just have to tweak the letter a bit.

This is dangerous. LOR for medical school should be medical school oriented. Of course it’s fine to use LORs for both AACOMAS and AMCAS. If the professional programs are different, make sure you ask them to tweak it. However, without ever knowing the content they might not tweak it in the way you want. The last thing you want is to apply to like PhD and have it say “john is going to make a great physician!”

Make sure they know and be transparent with them. Most professors i dealt with were willing to tweak a previous LOR for multiple programs/jobs

Your question is confusing me, partly because you asked it in the pre-DO forum. If you have professors who wrote letters for you for medical school, they only have to be uploaded through AMCAS and AACOMAS. Chances are you're going to want a different type of letter for non-clinical professional programs or other graduate programs. In that case, they will actually have to modify the letter they wrote. Their willingness to do so will depend on a number of things 1) how much they like you and feel like they can recommend you for these other programs, 2) how much notice you give them and 3) how busy they are. Your best bet is to ask them directly and give them an out. "Dear professor Smith, I really appreciate the letters you wrote for Medical school and XX. I have decided to also apply to (insert program) and was wondering if you would be willing to submit a recommendation on my behalf by (insert date). If this is not possible, please just let me know. I believe your recommendation is particularly relevant because of (reason), but I am confident I could fill in the gap with another letter if you are unable."

in general, letter writing is part of the professor gig, so they probably have a formula they use to make it less painful.
 
Yes they need to update the letters for each. The letter should be speak towards your suitability for that particular program/profession.
