When is the optimal time to send a letter of interest pre-II to schools and post-Interview?

In addition, regarding update letters, what are considered "good" updates. I have been lucky enough to publish 3 more papers (2 FA) and receive an award for my research at two conferences. Are these substantial enough updates or are there other things they might be looking for?

A letter of interest is worthless. Schools know you're interested because you sent them an app.
