Received glowing LOR from my direct supervisor (he has a PhD and his title is "research scientist"), but now I am hearing that it should've been PI?

As the title says...
I received an amazing LOR from my boss. I figured since I work with him I should ask HIM for the LOR rather than the PI, since I barely know the PI -- joined the lab during the pandemic. I've been out of undergrad two years now and I've been at this job for one year.

Do I need to ask the PI for a letter too now even though he doesn't know me much at all? My boss already submitted the letter to ve collect, so he can't have the PI just add his signature and resubmit it.

If a PI has PhDs in his lab working under him and you are working under the PhD, the PIs signature doesn't add much if this is just an application for an MD program (if it is MSTP that might be different).
