The MOA in FA for rifampin is a bit confusing. It says that it is a DNA dependent RNA polymerase inhibitor but then in the diagram for antivirals, rifampin is shown as blocking packaging and assembly of vaccinia. Does it do two things?

Yeah. The assembly of vaccinia is reversibly blocked by rifampin. I remember reading/hearing somewhere that mutant vaccinia strains, demonstrating rifampin-resistance, have D13L mutations.
