I recently updated my grades on OptomCAS. What I am asking is do I need to send in another copy of my official transcript to verify everything again? Thanks

YES and then they take 2 weeks (foreverforevERFOREVER) to do what they do
a.k.a. confirm the grades and THAN send it to the schools

Yes, you're correct. I called ICO today and they did confirm that you have to send the official transcripts again
