Lab Manager Job App: Can I send an email to the PI if one of my references just came through?

I recently applied to a job which I'm very excited about, and I could not reach one of my PIs for a reference. I put down the masters student as my supervisor who has a better idea of what I do anyways-- but it turns out he was just on vacation and is actually very happy to be a reference for me.

Can I email the PI of the lab I applied to with an additional reference and a 'hello'?
